Facility Features, Working Principle and Specifications
Facility Description
The UHPLC-HRMS Triple Quadrupole (Ultra-High-Performance Liquid Chromatography coupled with High-Resolution Mass Spectrometry using a Triple Quadrupole analyzer) is a sophisticated analytical instrument designed for highly sensitive and selective quantification of chemical compounds. It combines the advanced separation capabilities of UHPLC with the precision and reliability of triple quadrupole mass spectrometry.
Sample Injection & Separation:
The liquid sample is introduced into the UHPLC system, where it undergoes rapid and efficient chromatographic separation into individual components.
Ionization:The separated components are converted into charged ions by ionization techniques like HESI or APCI.
Mass Analysis in QQQ:Quadrupole mass analyzers contain four parallelly assembled metallic rods that act as electrodes. A high-frequency electric field is created by applying direct-current (dc) and radiofrequency (rf) potentials across these electrodes. The stable vibratory motion of ions is exploited to attain mass separation through the rf-dc. For a given set of rf-dc parameters, only certain ions within a narrow range of m/q would have established stable trajectories.
Data Analysis:The data is processed using software for quantification.
Resolution: 70,000 FWHM
Mass range:50 to 2000 amu
Ionization method: HESI, APCI
High mass accuracy (in the range of ppb)
Instructions for Registration, Sample Preparation, User Instructions and Precautionary Measures
An appointment will be given as per queue and will be informed by email.
At least 24 hours prior information should be given to ESED lab if users want to cancel/postpone the slot.
Sample preparation must be completed by the user.
The sample must be filtered using a 0.22 µm nylon filter.
Ensure that analyte concentrations are less than 3 ppm.
1. First discuss with the facility personal about the nature of sample and required analysis. Please proceed with payment only when you get a confirmation that the required analysis is feasible.
2. A requisition letter addressed to Head, ESED along with a printout of electronic fund transfer (NEFT) proof should be sent by post or submitted in person to ESED office, IIT Bombay, Powai, Mumbai-400076.
3. If you are sending samples by post please complete al details in Requisition form.
4. Once we receive the requisition form and advance payment, your appointment will be scheduled as per the queue.
5. The users will be informed about their date and time of slot by e-mail.
6. We prefer that you or your representative, who knows/understands the sample/material should be present on the day of appointment.
7. Before proceeding further, please review How to Use Facility and Payment Procedure for more information.
